When we arrive at the stop line of an intersection and there is already a car stopped there, do we need to stop again when it's our turn?

Yes, we must stop again at the stop.

If, on arriving at an intersection regulated by a Stop, whether as a road marking or as a vertical sign, we find another vehicle stopped, once it is our turn we must stop again to carry out the Stop correctly, looking both ways along the intersection and deciding the best moment to cross it. Under no circumstances should we follow the vehicle ahead of us without having stopped first.
